Effects of ionizing radiation on the STAT3 expression of gastric adenocarcinoma cells
This study is to investigate the effect of ionizing radiation on the activity and localization of STAT3 in gastric adenocarcinoma cells. After irradiated with ,60,Co γ-rays, the content and activity of STAT3 in gastric adenocarcinoma BGC-823 cells were detected by Western blot. The content of STAT3 in nucleus of BGC-823 was further detected by Western blot after the total protein in nucleus of irradiated BGC-823 was extracted. Finally, the distribution of p-STAT3,(Y705), in BGC-823 treated with or without irradiation was examined by immunofluorescence staining. The results showed that the level of p-STAT3,(Y705), in BGC-823 cells decreased significantly after ,60,Co γ-rays irradiation, and the content of STAT3 in BGC-823 cells nuclei also decreased after irradiation. As predicted, the p-STAT3 ,(Y705), localized in nucleus of BGC-823 cells also decreased significantly. ,60,Co γ-rays irradiation inhibited the phosphorylation of p-STAT3 ,(Y705), and further suppress the nucleus localization of STAT3, which might be involved in radiation-induced cell proliferation inhibition and apoptosis of gastric adenocarcinoma cells.
