实验以丙酮丁醇梭菌(,Clostridium acetobutylicum,)CICC 8012为出发菌株,采用高能脉冲电子束(High energy pulsed electron, HEPE)对丙酮丁醇梭菌进行辐照诱变,通过含丁醇的固体培养基对诱变菌株进行胁迫,诱使丙酮丁醇梭菌的丁醇耐受性提高。经过多次诱变和筛选,最终获得了一株诱变菌株S10,其丁醇产量为8.32 g/L,较出发菌株产量提高了10%。同时探讨了微量元素对诱变筛选菌株S10发酵的影响,在发酵培养基中添加醋酸铵、硫酸亚铁和硫酸锰等微量元素,考察诱变筛选菌株S10丁醇产量的变化。结果表明,发酵培养基中添加一定量醋酸铵、硫酸亚铁和硫酸锰等微量元素,分别可将诱变筛选菌株S10的丁醇产量提高至10.21g/L、9.13 g/L和9.21 g/L。
Abstract
This thesis takes ,Clostridium acetobutylicum, CICC 8012 as the original strain and uses HEPE(High energy pulsed electron) to mutate the strain. With progressive cultivation method, a new strain S10 was selected, which has a high production of butanol with 8.32 g/L, increasing 10% compared to the original strain. And then, the experiment of adding CH,3,CHOONH,4, FeSO,4, and MnSO,4, was carried out to explore if these factors had effect on the fermentation production of butanol. The result showed that these factors could increase the butanol production with S10.
