THE HYPERTHERMIA RADIOSENSITIZATION OF HUMAN GLIOMA CELLS SHG-44 AND THE EXPRESSION OF FAS/FASL
The effects of hyperthermia and radiation on human glioma SHG−44 cells and the alteration of Fas/FasL expression in the treated cells were investigated. The sensitivity of SHG−44 cells to hyperthermia and radiation was measured by colony forming assay and flow cytometry (FCM). The expression of Fas/FasL was detected by RT−PCR. By hyperthermia ( 42°C, 40min) combined irradiation, the colony forming ratio was reduced in a more effective way than by just the irradiation. The TER was 1.95. The apoptosis ratios were 20.2, 29.3, 59.1%, respectively, for the control group , the just-irradiation group and the hyperthermia combined radiation group. The expression of Fas was found in the three groups, but the expression of FasL could be found only in the two treatment groups. Hyperthermia could boost the effectiveness of radiation against glioma cells significantly. This also indicates some relationships between apoptosis and Fas/FasL.
