Selection of the respiration deficiency mutant yeast irritated by laser and optimization of the fermentation condition
A screen of respiration deficiency mutants of ,Saccharomyces Cerevisiae,YE0 induced by 266nm laser irradiation was studied. After the selective culture of TTC medium and determination of the alcohol yield by gas chromatographs, a respiration deficiency mutant JB7 with high glucose transformation efficiency was obtained. The results show that under the optimized fermentation conditions ( inoculation amount is 15%, glucose concentration is 25%, fermentation temperature is 32℃ and fermentation time is 72h) the alcoholic yield in volume fraction is 12.3% and the glucose transformation efficiency of JB7 is 52.9%, which are higher than that of the original strain YE0(glucose transformation efficiency is 49.6%, the alcoholic yield is 12.0%). The high glucose transformation efficiency shows a good prospect of application of the respiration deficiency yeast.
