CHANGES OF DNA POLYMERASE β GENE TRANSCRIPTION AND EXPRESSION IN HEPATOMA AFTER IRRADIATION
Upon the previous studies about the effect of radiation on the DNA polymerase β (pol β) activity and the roles of the enzyme on DNA repair synthesis, we reported the effects of γ-irradiation on gene transcription and its expression of DNA pol β. After 2 or 4 Gy total body irradiation, some biological properties of the enzyme in SMMC-LTNM hepatoma cells borne in nude mice changed. The content of DNA pol β mRNA in hepatoma cells increased significantly, reached the maximum (about 1.76-fold of control) at 36 hours, then decreased closely to control level 3 days later. The gene expression tested by immuno-gold-silver staining assay exhibited some alterations which showed that the enzyme gene expression become more active at 48 hours after irradiation. These results indicated that γ-irradiation would promote DNA pol β transcription and enhanced its DNA repair function.
