通过80%硫酸铵沉淀与Sephadex G-50凝胶过滤层析,从当归饮片中得到分子量为18.33 kDa的当归蛋白(命名AP)。用X射线对小鼠进行全身照射至吸收剂量6 Gy,建立急性照射损伤模型,以阿米福汀为对照,监测小鼠的体质量、外周血白细胞数、胸腺指数、脾脏指数、脾集落形成单位(CFU-S)、脾脏表观、脾组织病理分析和脾脏抗氧化活力等指标,检验当归蛋白AP的辐射防护效应。结果表明,当归蛋白AP可使受照小鼠的外周血白细胞数量、胸腺指数、脾脏指数、CFU-S和脾脏的过氧化氢酶活力分别增加135%、103%、23%、219%和180%,改善脾白髓损伤,使其体质量下降率减少94%。结果提示,当归蛋白AP对小鼠具有辐射防护作用。
Protein named AP with the molecular weight of 18.33 kDa was purified from prepared slices of ,Angelica sinensis, using 80% ammonium sulfate precipitation and Sephadex G-50 gel filtration chromatography. A mouse model of acute radiation injury was then established via whole-body irradiation with X-rays at a dose of 6 Gy. The radioprotective effects of ,Angelica sinensis, protein AP were evaluated using amifostine as a control by measuring body weight, peripheral blood leukocyte count, thymus index, spleen index, colony forming unit-spleen (CFU-S), morphology and histopathology of the spleen, and splenic antioxidant enzyme activity in the irradiated mice. The results showed that AP significantly increased the number of peripheral blood leukocytes, thymus index, spleen index, CFU-S, and catalase activity in irradiated mice by 135%, 103%, 23%, 219%, and 180%, respectively, and kept the number of splenic white pulp, resulting in a significant 94% reduction in weight loss. These results indicate that the ,Angelica sinensis, protein AP has radioprotective effects on mice.
