      摘要:A screen of respiration deficiency mutants of Saccharomyces CerevisiaeYE0 induced by 266nm laser irradiation was studied. After the selective culture of TTC medium and determination of the alcohol yield by gas chromatographs, a respiration deficiency mutant JB7 with high glucose transformation efficiency was obtained. The results show that under the optimized fermentation conditions ( inoculation amount is 15%, glucose concentration is 25%, fermentation temperature is 32℃ and fermentation time is 72h) the alcoholic yield in volume fraction is 12.3% and the glucose transformation efficiency of JB7 is 52.9%, which are higher than that of the original strain YE0(glucose transformation efficiency is 49.6%, the alcoholic yield is 12.0%). The high glucose transformation efficiency shows a good prospect of application of the respiration deficiency yeast.

      摘要:In order to investigate the effect of carbon ion irradiation on apoptosis and Bax/Bcl-2 expression in human tongue carcinoma cells, exponentially growing human tongue carcinoma cells (Tb) cultured in vitro were irradiated with 0, 0.5, 1.0, 2.0 or 4.0 Gy of 12C6+ ions respectively. Survival rate of irradiated cells at various doses were measured by MTT assay. The nucleus changes of apoptosis and necrosis of cells stained by Hochest/PI were observed through fluorescence microscope. The cell cycle changes were detected by flow cytometry (FCM). The expressions of Bax and Bcl-2 were detected by Western blot analysis. The results show that the viability of Tb cells decreases gradually with increment of irradiation doses of carbon ions. The proportions of apoptosis cells in the irradiated groups are significantly higher than those in the control group. There is a positive correlation between irradiation doses and retardation strength in G2 /M phase at 24 h after irradiation (P<0.05). And the expressions of Bax and bcl-2 are significantly up-regulated and down-regulated respectively by 12C6+ ion irradiation. It can be concluded from above that cell apoptosis induced by heavy ion with high-LET may be mediated through the Bax/Bcl-2 expression pathway.

      摘要:Cell apoptosis and the expression of DNA damage repair-related genes XRCC2, ATM and Lig4 were detected separately by flow cytometry analysis and real-time fluorescence quantitative (RT-PCR), in order to investigate the effect of Trichostatin A(TSA)pre-treatment on the radiosensitivity of esophageal cancer EC9706 cells to X ray in vitro. The results indicate that pre-irradiation exposure to TSA enhances cell apoptotic rate by X ray markedly (P<0.05) and the mRNA levels of XRCC2, ATM and Lig4(except 8Gy)are up-regulated by X ray individually(P<0.05). TSA alone and TSA in combination with X ray all reduce mRNA levels of XRCC2, ATM and Lig4. In conclusions, TSA may radio-sensitize EC9706 cells to X ray by reducing mRNA levels of XRCC2, ATM and Lig4.

      摘要:Reasonable segmentation of the individual comet contour from the Comet Assay (CA) images is the precondition for all of parameters analysis during the automatic analyzing for the CA. The Otsu method and several arithmetic operators for image segmentation, such as Sobel, Prewitt, Roberts and Canny were used to segment the comet contour, and characters of the CA images were analyzed firstly. And then the segmentation methods which had been adopted in the software for CA automatic analysis, such as the CASP, the TriTek CometScoreTM, were put forward and compared. At last, a two-step procedure for threshold calculation based on image-content analysis is adopted to segment the individual comet from the CA images, and several principles for the segmentation are put forward too.

      摘要:In order to investigate decorporating and detoxicating acute depleted uranium poison of flavonoids, the decorporation effect of flavonoids with catechol group, chelating agent of catechols amino carboxylic acid type (8102) and dimethyl sulfoxide (DMSO) were studied under DU exposure and Wistar rats as animal model. In 3d later after injecting DU with dose of 20 mg/kg and then injecting flavonoids with dose of 10 mg/kg in abdomen, the mortality, uranium contents, kidney function and pathological observation of kidney were investigated. It has been found that nearly 20% mortality occurs both in the T2 and in T1 groups meanwhile 30% mortality occurs in the control group. But death has not been observed in 8102 group. The concentration of excreted urine uranium increases and the level of serum uranium decreases when T2 used. The retention of uranium in kidney with T2 group also increases. Large reducing of urea nitrogen and creatinine in serum has been observed in 8102 and F1 groups, and reducing of creatinine has also been observed in F2, T1 and T2 groups. The serious injury of kidney has not been observed in T2 group. The protection efficiency can be listed as T2> 8102>T1>F1>QU. It has been revealed that flavonoid T2 is the most effective protector from the damage and poison of DU.

      摘要:In this article, some factors, such as ultrasound time, ultrasound power and starch concentration, affecting corn starch paste’s viscosity and morphology of starch particle are studied by use of ultrasound. The starch treated by ultrasound is used to prepare corn hydroxypropyl starch (HPS) with propylene oxide under microwave. Compared with conventional methods, ultrasonic-microwave method can improve HPS’s degree of molar substitution (MS) and sharply shorten reaction time. It is found that ultrasound can destruct surface morphology of starch particle, which can increase reaction efficiency as the particle becomes relax and easily to react with propylene oxide. HPS’s MS increases along with the reaction time and concentration of propylene oxide. With the increment of starch concentration and microwave power, the HPS’s MS increases at beginning and then decreases. The optimal parameters for HPS by use of ultrasonic-microwave irradiation are as followings: ultrasound power is 600 W, ultrasound-treated time is 25 min, microwave power is 100 W, microwave-treated time is 14 min, starch concentration is 35% and concentration of propylene oxide is 6.4%.

      摘要:A new type of chain extender for amide, named diacetyl ethylenediamine (DAED), was synthesized using microwave heating-induced reaction. Aromatic polyureas were synthesized through the reaction of 4,4’-diphenymethane diisocyanate with ethylenediamine (EDA) and DAED, respectively. The reaction rate was much lower when using DAED as chain extender than that of using EDA as chain extender. All these results indicate that DAED is an effective chain extender for preparation of spray polyurea system.

      摘要:The changing rules of 2-dodecylcyclobutanone according to different package styles and adding free radical scavenger in irradiated beef during storage were investigated. 2-dodecylcyclobutanone was extracted and separated by silica gel column and analyzed by capillary column gas chromatography-mass spectrometry (GC-MS). The results show that package with preventing from light can induce an appreciable lowering of the concentration of 2-dodecylcyclobutanone formed in irradiated beef, but preventing from oxygen does not have the same effect. The effects of different packaging atmosphere (N2, vacuum, and air) on the stability of 2-dodecylcyclobutanone are almost similar during storage. The concentration of 2-dodecylcyclobutanone can be decreased significantly by adding free radical scavengers, and it will be weakened, however, as the storage time extended.

      摘要:In order to determine the effect of misstep irradiation on the absorbed doses of irradiated products in the process with moving shuffle-dwell irradiation system the contribution ratio of absorbed doses had been measured at 44 irradiation location with corrugated paper as model of irradiated products. It has been revealed that the contribution of absorbed doses obviously comes from that at some special irradiation location with the irradiated dummy. And misstep irradiation at special irradiation location has large effect on the total absorbed doses. In this paper the reason, identification and protection of misstep irradiation has been discussed. And the predicated formula of effect on absorbed doses has been given.

      摘要:In order to investigated the current radiation environment situation of a mine from 2005 to 2007, various of radiation monitoring instruments have been used to measure the concentration of radon and its daughters, γ radiation level and natural radionuclide content in a mine including industrial site and living area respectively. The obtained data show that radon and its daughters concentration and γ radiation level in most of the monitoring industrial site is lower than the national management limits for the quality evaluation of radiation environment in nuclear industry. And in the living area, individual dose equivalent calculated after deducting the background at all monitoring point is also lower than the national management limits for the quality evaluation of radiation environment.

      摘要:The work is to explore the protection of magnesium sulfate(MgSO4) on radiation-induced acute brain injury. Thirty six mature Sprague-Dawley(SD) rats were randomly divided into 3 groups of control, experimental control and experimental therapy group. The whole brains of SD rats of experimental control and experimental therapy group were irradiated with a dose of 20 Gy using 6 MeV electron beam. MgSO4 was injected into the abdomen of experimental therapy rats group 1day before, immediately and continue for 5days after irradiation respectively. The brain tissues were taken on 3, 10, 17 and 24 d after irradiation. Ca2+ content in brain cell was measured by laser scanning confocal microscopy, and the NO content in brain tissue was detected by the method of nitric acid reductase. Compared with the blank control group, the contents of Ca2+ in brain cell and NO in brain tissue of the experimental control group increase (P<0.05). Compared with the experimental control group, the two indexes of the experimental therapy group decrease (P<0.05). It shows that MgSO4 used in early stage can inhibit the contents of Ca2+ in brain cell and NO in brain tissue after radiation-induced acute brain injury. It means that MgSO4 has a protective effect on radiation-induced acute brain injury.

      摘要:In order to study the peroxidation reaction of lenoleic acid induced by UV the dried thin film and so-lution prepared by linoleic acid were irradiated by UV light (254 nm). The production, MDA and diene conjugate, formed in peroxidation reaction of dried thin film and lenoleic acid solution was measured by thiobarbituric acid (TBA) reaction and absorption spectra. The results show that the Lipid-peroxidation of the dried thin film and lenoleic acid solution increases with the UV irradiation dose and decreases with UV irradiation dose rate. The dried thin film is more sensitive to the UV radiation than the solution.
