      摘要:In this paper the radioprotection effects of fullerene derivative (C60-Lys) on AHH-1 cells and its protective mechanism have been investigated. The 800 mg/L C60-lys can improve the survival rate of irradiated AHH-1 cells, which are 64.6% and 41.3% for the irradiation and control groups at 8 Gy dose, respectively. The cell fluorescence images show that the 16.3% apoptosis rate at 1200 mg/L C60-lys is lower than that of the 39.2% irradiation free of C60-lys. In the situation with additive 800 mg/L C60-lys and 4 Gy of irradiation, the dosed time before or after irradiation significantly affects the average survival rates of AHH-1 cells, which are 85.6% and 64.1%, respectively. In the AHH-1 cells treatment with C60-lys can obviously reduce the G2 blockage of AHH-1 cells induced by 4 Gy radiation, DNA damage and amount of 8-OHdG especially.

      摘要:In order to learn the effect of β-actin in acute radiation injury, the changeable pattern with time of plasma β-actin, PT, APTT, FIB and IL-8 in mice spleen tissue exposed to 6 Gy γ-rays radiation was investigated. Blood and spleen were collected at immediate, 1, 2, 3, 4, 7 and 14 d after irradiation, respectively. The contents of blood β-actin were detected by magnetic bead separation enzyme-linked immunosorbent. An STAGO blood coagulation instrument was used to determine PT, APTT and FIB. DNA expression of IL-8 was detected by real time-PCR analyzer. The results show that the level of β-actin in serum of irradiated mice is higher than that of normal control group at all different post-irradiation time points although the change of β-actin in serum of irradiated mice with time schedule shows a pattern which increases within 1d and declines beyond 1d. The trend of the changes in plasma PT, APTT, FIB and in spleen IL-8 and time pattern of these changes are similar to that in plasma β-actin in irradiated mice. The difference in values and the time phase between plasma β-actin and other indexes is the reaching time of peak values and the declining levels of the values. These results are valuable for studying the role of β-actin in acute radiation sickness pathology process and can be used to explore new factors influencing and regulating pathology process.

      摘要:In order to establish the stable clones of RAW264.7 cells with S100A8 gene overexpression, and directly investigate the role of S100A8 in γ-rays induced hematoiesis/immune system injury, RT-PCR method and immuno-fluorescence staining analysis were performed to measure the expression of S100A8 mRNA and protein. The cell cycle was detected by flow cytometry at 6 h after irradiation with 10 Gy γ-rays. The level of reactive oxygen intermediates (ROIs) was detected by flow cytometry at 6h after H2O2 treatment. The cell cycle analysis showed that the percentages of RAW264.7/S100A8 cells had an increase in G0/G1 phase and a decrease in S phase, and G2/M arrest didn’t occur in experimental group while the level of ROIs was lower than that of the negative control cells. These results indicate that radiation resistant of RAW264.7/S100A8 cells may be related to the antioxidant role of over-expressed S100A8 in comparison to RAW264.7/pcDNA3.1+ cells.

      摘要:To study the effects of ELAC2 on the invasion, migration and radio-sensitivity of human breast cancer cell MCF-7, ELAC2 genes were transfected into MCF-7 using Lipofectamine transfection assay. Western blot assay was used to detect the protein expression of ELAC2, cyclinB, CDC2 and P53 while wound healing assay and Boyden chamber assay were performed to determine cell invasion and migration, and colony formation assay and flow cytometry assay were used to detect cell surviving fraction and cell cycle. The results show that MCF-7/ELAC2 cell lines with overpression of ELAC2 were successfully established. The overexpression of ELAC2 could obviously inhibit invasion and migration of MCF-7 cells. The overexpression of ELAC2 enhanced the radiosensitivity of MCF-7 cells after x-ray exposure and caused increased ratio of G2 phase. At the same time cyclinB protein expression decreased and the expressions of P53 and Tyr-15 phosphorylated CDC2 up-regulated. The results suggest that the overexpression of ELAC2 may obviously inhibit invasion and migration of MCF-7 cells, and may result in radiosensitivity enhancement of MCF-7 cells to X ray, which could be associated with either down-regulation of protein expression of cyclinB or up-regulations of protein P53 and Tyr-15 phosphorylated CDC2.

      摘要:Recently antibiotics wastewater in environment has induced the increment of bacterial resistance. This paper is to investigate the gamma radiolysis of wastewater containing 10 μg/mL levofloxacin lactate (LVF). It has been found that the antibiotic was removed more than 99% with 1 kGy under air while the G-value decreased with the dose increment. Five main degraded products (m/z 346, 330, 318, 302, 274) and the most probable radiolysis pathway were identified by liquid chromatography-mass spectrometry (LC-MS). Compared with the degradation under N2, the radiolytical mechanism was suggested. In the active assay, 2 μg/mL was the inhibitory concentration. Compared with the concentration of 4 μg/mL, the minimal inhibitory concentration (MIC), degraded products analysis can be suggested that the byproduct m/z 346 might have the anti- E. coli activity. It has been shown that 3 kGy is the appropriate dose for the radiolytical treatment of LVF. To sum up, the gamma radiation technique is an effective method for decomposing antibiotics, and it is necessary to take the activity of degraded products into consideration.

      摘要:Pulse radiolysis experiments were conducted on the probe molecule, pyrene, in acetonitrile, acetone, cyclohexane, ethanol and mixed solutions containing ammonium ionic liquid with different volume. The formation of pyrene radical anion (Py·-) was observed at 490nm and the rate constants of the Py·- formation were determined in the ethanol solution and in the mixture solution with different ionic liquid volume fractions, respectively. Moreover, the rate constants are changed greatly with increasing of the ILs concentration in the mixture solutions. The results can be assumed that the dry electrons produce in very short time scales after electron pulse and the solvated electrons can react with pyrene to generate its radical anion in nanosecond time scale.

      摘要:In order to investigate the radioprotection property of water-soluble chitosan with low-molecular weight, sodium hydroxide eluting and oxidation degradation of chitosan with H2O2 under HAc condition were used to prepare water-soluble low-molecular weight chitosan. MTT assay and flow cytometry (FCM) assay were performed to evaluate the cell survival rate, apoptosis and cell cycle alteration, respectively, after γ-rays irradiation. The results show that water-soluble low-molecular weight chitosan was successfully prepared, and the degree of deacetylation (D.D) of chitosan was increased from 79.3% to 94.1% while the molecular weight was decreased from 8.50×105 to 3.26×103 after treatment for 120 min. Low-molecular weight chitosan (LMWC) has good radio-protection effect to BRL cells from radiation damage induced by 4 Gy 60Co γ-rays in evidence of higher survival rate and lower SubG1 peek of the cells. After treatment with 50 μg/mL of LMWC, survival rate of BRL cells increased from 66.5%±0.19% to 87.5%±0.16% at 48 h (p<0.05), the peak of SubG1 decreased from 51.9%±2.1% to 16.9%±1.3% at 24 h after 4 Gy 60Co γ-irradiation (p<0.05).The results indicate that low molecular weight has excellent radio-protection effect to normal rat liver cell line (BRL).

      摘要:A new hydrogel film was prepared by use of poly (vinyl alcohol) (PVA), poly(ethylene oxide)(PEO) and chitosan under γ-rays irradiation. The influences of absorbed dose and the chitosan content on the properties of hydrogel film were investigated. The results showed that the gel fraction increased with the absorbed dose increasing, but the swelling ratio decreased. The gel fraction could reach 70% when the absorbed dose up to 21 kGy. The gel fraction, tensile strength and elongation of the hydrogel films reduced with the increment of absorbed dose when it was more than 21 kGy. With the increment of chitosan content, the gel fraction and tensile strength as well as elongation of the hydrogel films decreased, but the swelling ratio increased. The special functional groups of chitosan didn’t change after irradiation, and the antimicrobial activity would be retained. A new kind of hydrogel film was obtained.

      摘要:In order to investigate microstructure and chemical composition changes of titanium alloys by intense pulsed ion beam radiation, titanium alloys were bombarded by intense pulsed ion beams(70%H+ and 30%C+)with energy density of 4.5 J/cm2.The microstructure and chemical compositions of cross-section of samples irradiated by ion beams were investigated with scanning electron microscope. The results show that titanium alloy samples are composed of two phases, belonging to the type of α+β titanium alloy. The microstructure is different in different areas; there are the same types of microstructure in most of areas. In some local areas, it is typical clostridium organization. The microstructures of titanium alloy irradiated by intense pulsed ion beams were changed with depth of tens of microns. The β-phase structure was damaged to some extent with the range of depth. The change in microstructure of titanium alloy irradiated by intense pulsed ion beams is due to thermal stress caused by surface temperature gradients and its spread in the target.

      摘要:The effects of γ-rays, electron and proton irradiation on two different types of GMR (Giant magneto resistance) multi-films containing different materials were investigated. Lower than 10% decrement in MR rate were observed in all the irradiation process meanwhile the coercivity and exchange field had not varied. The proton irradiation as the major irradiation in the space has much less affection on GMR element than the other two irradiations. This good performance makes GMR element having further application in space science.

      摘要:By the study of post-radiation and accelerate annealing in the total ionizing dose(TID), we find out that the additional 50% dose radiation and high temperature accelerate annealing (100℃ 168h) can not be skipped in the test because it is necessary to strictly check the memory’s TID effect in the space radiation environment. The analysis of device's discreteness in the test shows that parameters of device should be tested before the radiation to get their standard deviation. The high reliability of the memorizer which used in the satellite can be ensured by rejecting the device which increases the standard deviation. The TID test in the NASA report have been confirmed that the effectiveness of parameter test in 'read-only’, 'write’ and 'erase’ mode is better than the test in 'only read’ mode as it can reflect the degradation characteristics of the memorizer following ionizing radiation more reasonable. The test sequence applied for each memory at any electrical measurement steps after exposures annealing should be considered.

      摘要:By use of fuzzy subtractive clustering model, the relationship between tensile strength of radiation vulcanization of NBRL (Nitrile-butadiene rubber latex) and irradiation parameters have been investigated. The correlation coefficient was calculated to be 0.8222 in the comparison of experimental data to the predicted data. It was obvious that fuzzy model identification method is not only high precision with small computation, but also easy to be used. It can directly supply the evolution of tensile strength of NBR by fuzzy modeling method in radiation vulcanization process for nitrile-butadiene rubber.

      摘要:To observe the clinical efficacy and adverse reactions of skin cancer treated with heavy ion beams(12C6+), 12 patients (volunteers) accepted the clinical therapy. The heavy ion beam energy was 100 MeV/u, RBE (Relative biologic effectiveness) value was 2.5—3.0, and radiation absorbed dose was 60—70 GyE for 6—7 d with 2.5—5.41 Gy/d. Clinical efficacy was valuated by tumor fashion in 3 months after treatment and acute adverse reactions judged by radiation therapy oncology group based on criteria of acute radiation injury. 12 patients were followed up for 3 months and detected. Among them 10 patients were completely remitted (CR) and 2 partly remitted (PR). No adverse reaction appeared except that grade 1—2 skin side-effect was observed. It is suggested that heavy ion beams are effective and safe treatment rays to skin cancer. It can shorten treatment time markedly and has no serious adverse reactions.
