      摘要:The human umbilical vein endothelial cells (HUVECs) of logarithmic growth phase were divided into three groups for different treatment. And then they were detected with the cell cycle distribution by flow cytometry, rate of apoptosis by annexin V/PI double staining method, expression of Bcl-2 protein by Western blot and expression of Bcl-2 mRNA by RT-PCR. The results showed that magnesium sulfate could advance the G2/M phase block of HUVECs after X-ray irradiation for 24 h, and improve the sensitivity of HUVECs to X-rays after irradiation for 24 h, 48 h and 72 h, increasing apoptosis rate to a certain extent (t=4.24, 7.19, 3.20, p<0.05) and decreasing Bcl-2 mRNA expression (t=3.034, 6.182, 3.957, p<0.05)after irradiation for 24 h, 48 h and 72 h, as well as inhibit the anti-apoptotic protein Bcl-2 protein expression significantly after irradiation for 48 h and 72 h. Therefore, magnesium sulfate could improve the G2/M phase arrest, increase the apoptosis rate and reduce Bcl-2 expression of HUVECs irradiated with X-rays.

      摘要:The data of dose verification for head tumor in the treatment of volumetric modulated arc therapy (VMAT) was compared to analyze whether or not the statistics difference of gamma passing rates exists when the secondary interlocking occurs. The verification planned for head tumor of 50 patients were used for testing and verifying with the chamber array detector from PTW(PTW Detector 729). Five comparative experiments were performed for each pair of the three standards. The gamma passing rate of 1%/1 mm, 2%/2 mm, 3%/3 mm and T-test were used in the quantitative analysis. Statistical differences were found for all measurements of the three standards paired groups (t value range: 12.647~62.911, p<0.001). The comparative analysis showed that the gamma passing rates was affected significantly when the secondary interlocking occurred. It is suggested that the treatment should be stopped until troubleshooting when the secondary interlocking occurred frequently in the radiotherapy.

      摘要:The aim is to observe the protective effects of plant lectin PHA-L against radiation injuries and the relative mechanism. 30 d survival experiment was used to observe the survival condition of mice 30 d after irradiation with 7.2 Gy γ-rays. Blood and immunological experiments of mice were carried out to observe WBC (White blood cell), femoral nucleated cells, bone marrow DNA content and organs index after irradiation with 7.2 Gy γ-rays. Intestine pathological section of mice irradiated on abdomen with 9.0 Gy γ-rays was prepared to observe the pathology of intestine. The results showed that compared with the irradiation group, the survival rate of mice treated with PHA-L increased, especially for the high dose group (60% lifted); compared with the irradiation group, white blood cell count, bone marrow DNA content and spleen nodules of mice administered with high dose group increased from 0.74±0.16, 1.02±0.17 and 9.80±6.46 to 1.18±0.40, 1.22±0.17 and 18.10±6.87 respectively, which were statistically significant (p<0.05). Moreover, PHA-L also had some protective effect on the organ. In pathology observation, the intestines of mice treated with PHA-L recovered significantly as well. Therefore, plant lectin PHA-L can protect mice from radiation injuries, yet the mechanism needs further study.

      摘要:Human umbilical vein endothelial cells (HUVECs) were irradiated with different doses (10, 20 and 40 J/m2) to investigate the role of JAK-STAT pathway and IL-6, IL-8 in UVB irradiation. After 24 h, the expression level of IL-6 mRNA and IL-8 mRNA was detected by real-time fluorescent quantitative PCR, and that of JAK1, JAK2, STAT1, STAT3, p-JAK1, p-JAK2, p-STAT1 and p-STAT3 was measured by Western-blot. Compared with the control group, the expression level of IL-6 mRNA and IL-8 mRNA increased, and that of JAK1, STAT1, p-JAK1, p-STAT1, p-JAK2 and p-STAT3 increased with the dose of UVB, except JAK2 and STAT3. It can be concluded that JAK-STAT pathway and IL-6 together with IL-8 play a role in HUVECs with UVB irradiation.

      摘要:The aim is to investigate radiation protection effect induced by carboxymethyl-β-1,3-glucan on Human B lymphoblasts(HMy2.CIR). The survival rate of carboxymethyl-β-1,3-glucan on HMy2.CIR was tested by CCK-8 assay at concentrations of 0, 0.01, 0.1, 1, 100, 200 and 400 μg/mL, respectively. The flow cytometry was used to measure the apoptosis rate. Micronucleus rate was assessed by the in vitro micronucleus test. DNA strand breaks were assessed by the comet assay. The results showed that treatment with carboxymethyl-β-1,3-glucan was not cytotoxic. Pre-treatment with carboxymethyl-β-1,3-glucan at concentration of 0.1 μg/mL for 72 h protected HMy2.CIR cells against radiation, as indicated by increased surviving fraction, reduced apoptosis, reduced micronucleus rate and fewer DNA strand breaks. The carboxymethyl-β-1,3-glucan has radiation protection effect on HMy2.CIR cells, which may be related to inhibition of apoptosis and DNA damage after radiation.

      摘要:To construct fluorescence expression plasmids pEGFP-N1-pprA and pDsRed1-N1-flag-pprI, pGADT-7-pprA and pet28a-pprI plasmid constructed at an earlier laboratory stage was used as the template, and Lipofectamine 2000 was employed to transfect two recombinant vectors into 293T cells. A fluorescent microscope was used for Fluorescence observation, and Western blot was employed to examine the expression of the fusion protein. Double digestions and the agarose gel electrophoresis showed that target bands appeared at 4 700 bp and 1000 bp, 4800bp and 1 100 bp. No apparent frameshift mutation occurred as shown in the sequencing results. The fluorescent microscopy showed red and green phosphors; the Western blot results indicated protein expression at 65 kDa and 60 kDa. pDsRed1-N1-flag-pprI and pEGFP-N1-pprA were successfully constructed to express proteins for eukaryotic cells 293T in vitro. The results indicated that prokaryotic genes pprA and pprI could co-express proteins in eukaryotic cells successfully, and laid a foundation for the interaction and synergism of pprA, pprI, and its products in the regulation network of radiation verified by DR, enhancing the radiation resistance of eukaryotic cells.

      摘要:The aim is to observe the protective effect of Vam3 on irradiation-induced injury of thymocytes isolated sterilely from mice. The thymocytes were divided into 6 groups: control group, resveratrol group, Vam3 group, irradiation group, irradiation with resveratrol group and irradiation with Vam3 group. Except the control group, resveratrol group and Vam3 group, the rest groups were exposed to radiation at a single dose of 4 Gy γ-rays from 137Cs after 30 min treatment by vehicle and tested drug, respectively. The thymocytes' viability was measured by bioluminescence; the level of thymocytes' reactive oxygen species (ROS) was measured by DCFH-DA; the level of thymocytes' γ-H2AX was measured by fluorescent antibody labeling; the early apoptosis was detected by FITC-Annexin V and PI labelling using flow cytometry (FCM) after radiation. Compared with control group, the thymocytes' cell viability decreased obviously after irradiation, and the level of cell ROS, γ-H2AX and early apoptosis cell increased. By comparing the resveratrol group and Vam3 group with the irradiation group, we can find that Res and Vam3 could reduce the decrease of thymocytes' cell viability and inhibit the increase of ROS, γ-H2AX and early apoptosis cell induced by irradiation. The comparison between irradiation with resveratrol group and irradiation with Vam3 group showed that the effects of Vam3 in increasing cell viability, decreasing the level of ROS and γ-H2AX and lowering the early apoptosis rate were more obvious than resveratrol. It can be concluded that Vam3 performs better than resveratrol in the protection of mice thymocytes against the acute radiation injury.

      摘要:105 keV Ti ions were implanted into SiO2 substrate, and were then subjected to thermal annealing in oxygen atmosphere. The formation, structure, distribution, optical absorption property as well as photocatalytic performance of the formed nanoparticles (NPs) were characterized using various techniques such as ultraviolet-visible spectroscopy, grazing incidence X-ray diffraction spectroscopy, transmission electron microscopy and atomic force microscopy. The results clearly show that high-fluence Ti ion implantation into SiO2 along with thermal treatments in oxygen atmosphere could cause formation of TiO2 NPs, which mainly present rutile phase. Morphology of the synthesized TiO2 NPs depends strongly on the applied fluence of Ti ions. With the fluence increasing, the irregular and isolated NPs could be transferred into densely distributed TiO2 NPs, resulting in formation of particle-like thin films. In addition, the synthesized TiO2 NPs at higher fluence show a certain photocatalytic degradation performance on Rhodamine B solution.

      摘要:To develop a method for rapid separation of low level 90Sr using magnetic particles, Fe3O4 nanoparticles were prepared by coprecipitation method, and poly (styrene/divinylbenzene) magnetic particles (Pst-DVB@Fe3O4, PF) were synthesized by emulsion polymerization. Finally a new kind of core-shell structure magnetic particle coated with DtBuCH18C6 (DtBuCH18C6@Pst-DVB@Fe3O4, DPF) was developed. The morphology, structure, composition and content of surface elements were analyzed by scanning electron microscope, FT-IR spectroscopy and X-ray photoelectron spectroscopy, etc. The batch experiment results for the separation of Sr2+ in aqueous indicated that the adsorption of Sr2+ to DPF increased with pH value and showed a drastic increase from pH 11 to pH 13. The kinetic model showed that the adsorption of Sr2+ to DPF was most likely to be a monolayer chemisorption process via forming complexation between DPF and Sr2+, which was consistent with Langmuir isotherm model. The maximum adsorption capacity of Sr2+ to DPF was 2.62 mg/g under optional conditions.

      摘要:In order to predict the dispersion of radionuclides both in atmosphere and aquatorium, a new model was established to couple the Gaussian puff model and Eulerian model. The results of the coupling model presented good agreements with the references, which demonstrated the validity of all the modules. The coupling model was applied to simulate diffusion and concentration distribution of 131I in atmospheric and aquatic medium in the fuel assembly meltdown accident of China lead-based experiment reactor. The results showed that the distribution range of atmospheric pollutions was within 10 km 2 h after accident, meanwhile distribution range of aquatic pollutions was between 10 km and 20 km. Under the fuel assembly meltdown accident, the contaminant concentration in atmospheric and aquatic are still below the national standard limit.
