研究杜仲叶浸提物对碳离子束辐照小鼠急性损伤的防护效应及其可能机制。小鼠灌胃不同剂量杜仲叶浸提物7 d后,6 Gy 12C6+束全身均匀辐照,照射后12 h处死小鼠,取其肝、肺组织,分别用黄嘌呤氧化酶法和硫代巴比妥酸法(TBA法)检测组织中的SOD活性和MDA含量;用中性单细胞凝胶电泳技术(亦称彗星试验)检测组织细胞的DNA损伤;并用流式细胞术检测组织细胞的凋亡率。和单纯辐照组相比,预先灌服不同剂量的杜仲叶浸提物, 能有效抑制重离子束辐照引起的小鼠肺和肝组织SOD活性降低和MDA含量提高;同时,还能使辐照小鼠肺和肝组织细胞的拖尾率、拖尾长度和细胞凋亡率显著降低,且有剂量效应关系。杜仲叶浸提物能显著减轻肝肺细胞DNA损伤和凋亡,推测这与其显著消除自由基作用有关。
The aim of this study was to explore the protective effects and possible mechanism of Eucommia ulmoides Oliver leaves extraction on acute injury induced by carbon ion beams in mice. The Kun-Ming mice were fed orally with crude extracts of Eucommia ulmoides Oliver leaves in various doses for 7 days, whole bodys were then irradiated with 6Gy 12C6+ ions, the liver and lung tissues of mice were collected at 12 h after irradiation. The activity of superoxide dismutase (SOD) and the content of Malondiadehyde (MDA) in liver and lung tissues were detected by the xanthine oxidase method and the thiobarbituric acid (TBA) reaction chromometry, respectively. Meanwhile, the DNA injury was determined by single cell gel electrophoresis (SCGE, comet assay) and the apoptosis was assayed by flow cytometry method. The results showed that there were dose-related significant decreases in MDA level, percentages of apoptosis cell, tail moment rate of cells and tail length in each concentration groups of drug. The dose-dependent marked increment of SOD activity in pretreated with drug groups compared with irradiation only groups. The extraction of Eucommia ulmoides Oliver leaves could greatly reduce the DNA danage and inhibite the cells apoptosis of irradiated mice. It can be deduced that the extraction of Eucommia ulmoides Oliver leaves can protect mice from the radiation damage due to its dramatic capacity scavenging free radical.Cited
