利用低能离子束生物技术对花生四烯酸产生菌(Mortierella alpina)进行诱变选育。研究表明,在离子注入(10keV,3×10,14,N,+,/cm,2,)条件下,后代菌株离散程度明显高于自然分离。经连续诱变处理,最终获得一株花生四烯酸高产菌I,49,−N,18,,该菌每升培养液可得生物量26.3g,干菌体中油脂含量为33.8%,其中花生四烯酸的含量占总脂的52.36%。而其AA产量高达4.66g/L,比对照N,7,菌株产量提高126.2%,且继代遗传功能稳定,表明I,49,−N,18,是一株极具工业化前景的高产菌,同时证明离子注入是一种有效的诱变手段。
Low energy ion implantation technology was used in mutation breeding on arachidonic acid (5, 8, 11, 14−eicosatetraenoic acid, AA) yielding starting strain, Mortierelle alpina N,7,. The results indicate that dispersion of descendants of the N,7, strain implanted with 10keV N,+, (3×10,14, N,+,/cm,2,) was bigger than that in natural division strain. I,49,−N,18, a high−yield AA-producing strain, was screened out by continuous mutagenicity. The biomass, lipid in biomass, AA in lipids of the high−yield strain were 26.3g/L, 33.8%(w/w), and 52.36%(w/w), respectively. The AA content in culture was 4.66g/L, which is 126.2% higher than the control, and its descendiblity was stable. It is concluded that I,49,−N,18 ,is a promising strain for industrialization, and that ion implantation has remarkable mutagenic effect on microorganism.
