To explore the molecular mechanism of G,2, arrest of EL-4 cells induced by X-irradiation, flow cytometry (FCM) and semi-quantity RT-PCR methods were used to observe the time-course changes of cell cycle and the mRNA levels of cyclin B1 and cdc2 of EL-4 cells after 4Gy X-rays irradiation. Results showed that the positive cells in G,2,/M increased at 2h and reached the maximum at 4—8h after the irradiation (,p,<,0.001). The cyclin B1 mRNA level of El-4 cells began decrease at 1h, reached the minimum at 24h after the irradiation. Meanwhile, the cdc2 mRNA level also declined during 12h to 48h, reduced to 59.2 %and 58.9% compared with that of control group respectively, and recovered at 72h after the irradiation. The results suggests that MPF(M phase promoting factor) activity reduction resulted from inhibition of cdc2 and cyclin B play a critical role in G,2, arrest induced by X-ray irradiation in EL-4 cells.
