探讨谷胱甘肽过氧化物酶(GPX)模拟物-环糊精(2-Se-CD)对中波紫外线损伤的防护作用。以小鼠成纤维细胞(NIH3T3)制备紫外线损伤模型,采用荧光分光光度法测定DNA裂解率,改良硫代巴比妥酸荧光法测定细胞脂质过氧化作用,流式细胞术测定H,2,O,2,。1.5kJ m,-2,紫外线(UVB)照射后24h可引起NIH3T3细胞 DNA裂解率、脂质过氧化、H,2,O,2,产生增高,2-Se-CD 可使前述变化发生不同程度逆转,即与紫外线损伤模型组相比, DNA裂解率、脂质过氧化、H,2,O,2,产生降低(,p,<,0.05—0.01)。结果提示,本实验室合成的GPX模拟物2-Se-CD具有防护紫外线损伤的作用。
The work was to study protective effect of 2-selenium Bridged β-cyclodextrin (2-Se-CD), a GPX mimic, on the damages induced by moderate-wave ultraviolet rays. UVB damage model was established with murine fibroblast, NIH3T3. DAPI and MDA were used to evaluate DNA lysis rate and lipid peroxidation level respectively, Flow cytometry was used to detect production of H,2,O,2,. Significant increases were found in the DNA lysis rate, the lipid peroxidation level and the H,2,O,2, production 24 h after the treatment of 1.5 kJ m,-2, UVB. However, adding 2-Se-CD to the sample solutions could reduce all the changes to different extent. The DNA lysis rate, lipid peroxidation level and the H,2,O,2, production decreased as compared with the damage model group (,p,<,0.05—0.01). The results suggested that 2-Se-CD might protect NIH3T3 from the damage induced by moderate-wave ultraviolet rays
