采用MTT方法和脉冲场凝胶电泳方法,以人肝L02细胞株为研究对象,对没食子儿茶素没食子酸脂(epigallocatechin gallate,EGCG)的辐射保护作用进行了研究。MTT结果表明,EGCG在5—50μmol/L的浓度下,对细胞有明显的保护作用,并对细胞的修复有促进作用;脉冲场凝胶电泳方法结果显示,EGCG作用可减少辐射所致的DNA双链断裂水平。两者均显示EGCG可减少对辐射导致的细胞死亡。
The radioprotecive effects of a major component of green tea polyphenone,(—)epigallocatechin gallate (EGCG) was examined by using the radiation — induced cell death model in Human — derived liver L02 cells,with the methods of MTT and pulsed field gel electrophoresis (PFGE). EGCG substantially suppressed the radiation — induced mortality under treatment concentration of 5—50μmol/L, where the cytotoxicity was mild. Significant suppression of DNA double strand break (DSB) was also observed with treatment of cells with 50μmol/L of EGCG. Both treatment prior to and behind irradiation was effective. It could be suggested that EGCG could either scavenge OH• or repair the damage of DNA of L02cells effectively.
