利用体外细胞集落形成法研究二氢丹参酮Ⅰ对肿瘤细胞放射敏感性的影响。实验表明,二氢丹参酮Ⅰ对人肝癌细胞株(QGY-7703)和人肺腺癌细胞株(SPC-A-1)有较强的细胞毒作用,半致死剂量LD,50,分别为8.86,μ,mol/L和7.73,μ,mol/L。该药物与X射线联合作用能增强细胞对射线的敏感性,1,μ,mol/L的二氢丹参酮Ⅰ作用于有氧细胞1h后照射,对QGY-7703细胞和SPC-A-1细胞的增敏比(SER)分别为1.42和1.62;QGY-7703细胞和SPC-A-1细胞的氧增比(OER)分别为2.08和3.03。1,μ,mol/L的二氢丹参酮Ⅰ在乏氧状态下对这两种细胞亦有放射增敏作用,QGY-7703细胞的SER仍为1.42,而SPC-A-1细胞的SER增大为1.96。在分次照射实验中,1,μ,mol/L的二氢丹参酮Ⅰ能完全抑制这两种细胞的亚致死性损伤修复(SLDR)。
In the experiments, the radiosensitization of dihydrotanshinone Ⅰ on human hepatocelluar carcinoma cells QGY-7703 and lung adenocarcinoma cells SPC-A-1 was studied through in vitro colony formation method. The results showed that dihytanshinone Ⅰ has cytotoxity to the two kinds of cells and the LD,50, was 7.73,μ,mol/L and 8.86,μ,mol/L, respectively. The compound enhanced the radiation effect. Under aerobic condition the SER (sensitizing ehnancement ratio) of the compound was 1.42 for QGY-7703 cells and 1.62 for SPC-A-1 cells at the concentration of 1,μ,mol/L. The OER (Oxygen enhancemen tratio) for QGY-7703 cells and SPC-A-1 cells was 2.08 and 3.03,respectively. Under hypoxic condition dihydrotanshinone Ⅰ also had radiosensitivity for the two kinds of cells, and the SER of SPC-A-1 cells increased to 1.96 while the SER of QGY-7703 kept 1.42. In split-dose radiation experiment,dihydrotanshinone Ⅰ could inhibit SLDR (sublethal damage repair) obviously at the concentration of 1,μ,mol/L.
