抗辐射菌,Deinococcus radiodurans,具有显著的DNA修复能力, 包括对丝裂霉素 (MC) ,紫外线 (UV) 及γ射线. 通过化学诱导野生型菌株KD8301而得到的突变体KH3111,对MC,UV及γ射线敏感, 对链霉素 (Sm) 具有抗性. KH3111的辐射抗性能够通过转染野生型抗辐射菌KD8301基因组中一个已被克隆的DNA片断而得到恢复. 转染实验表明, 突变体具有很好的自然转化能力, 即不缺乏重组修复能力. 本研究确定了KH3111内发生突变的基因及其核苷酸序列, 该基因 (,orf,144,b,) 所编码的蛋白质由284个氨基酸组成, 与其它已知的蛋白质不具有同源性, 即它是一种新的蛋白质. 通过DNA测序分析, KD8301和KH3111的,orf,144,b,序列只是一个碱基的改变, 即由G突变为A, 所编码的第149位氨基酸由甘氨酸变为谷氨酰胺. 说明甘氨酸是DNA修复基因,orf,144,b,所编码的蛋白质中的一个重要残基. 用KD8301的,orf,144,b,基因转染KH3111,得到转染体KH3112,其对MC,UV及γ射线都具有同母本 (KD8301) 一样的抗性. 在抗辐射菌中, 这种对多种DNA损伤剂产生抗性所需要的基因 (,orf,144,b,) ,我们命名为pprA (,Pleiotropic gene Promoting DNA Repair,) 基因.
Deinococcus radiodurans(Dr), possesses a prominent ability to repair DNA injury induced by various DNA-damaging agents including mitomycin C (MC),ultraviolet light (UV) and ionizing radiation. A DNA repair mutant Dr KH3111 is a streptomycin resistant (Sm,R,) derivative of KH311 which was generated by treatment with nitrosoguanidine and was sensitive to MC,UV and γ-ray irradiation. DNA damage resistance was restored in KH3111 by transforming with DNA of a cosmid clone pDC144 constructed from genomic DNA of Dr KD8301. The high transformation efficiency of KH3111 with DNAs of pDC144 derivatives indicates that the mutant is proficient in natural transformation competence,suggesting that recombinational repair is not defective in the mutant. Gene affected by a mutation in the mutant was identified and its nucleotide sequence was determined. A complete open reading frame (ORF) which encompassed the KH3111 mutation region was found and tentatively designated ,orf,144,b,. The deduced amino acid (aa) sequence of ,orf,144,b, consists of 284 aa and has no significant homology to other known proteins. The exact KH3111 mutation site was identified by sequencing DNA fragments amplified from genomic DNA of the mutant. One nucleotide was altered (G to A) in the sequence of ,orf,144,b, in the mutant. The KH3111 mutation causes the substitution of Gly for Glu at aa position 149 of Orf144b. It appears that the Gly substituted by this mutation would be one of the important residues to fulfill the function involved in DNA repair in Orf144b. Survival measurements of a revertant KH3112 which was generated by transforming with DNA of a polymerase chain reaction (PCR) product containing a part of the ,orf,144,b, gene of KD8301 showed that the resistances to MC,UV and γ-ray in the revertant were fully restored at equal level with wild type,indicating that the characteristic sensitive phenotype of KH3111 can be accounted for by the point mutation in the novel gene. Thus,the ,orf,144,b, gene required for the multiple-DNA-damaging agent resistance of Dr was designated pprA (pleiotropic gene promoting DNA repair).
