研究WR-2721(氨基硫醇类药)对放烧复合伤的预防效果及其对造血功能的保护作用。结果证明:WR-2721能明显提高放烧小鼠的活存率,并延长死亡动物的活存时间。用WR-2721预防后,放烧小鼠的LD50由对照组的5.19Gy增大到9.53Gy, DRF为1.84。WR-2721对放烧小鼠的预防作用以200mg/kg照射前立即给药效果最好。放烧小鼠用WR-2721预防后,与对照组相比,外周血白细胞数和骨髓有核细胞数下降幅度少,恢复快,骨髓粒单系祖细胞伤后第1d和7d及骨髓造血干细胞伤后第7d和14d明显高于对照组。说明,WR-2721对放烧小鼠的预防作用,至少部分是通过保护造血功能而实现的。
Protective effect of agent WR-2721 on hematopoiesis and prophylaxis for the combined radiation-burn injury were investigated in the present study. It was found that pretreatment with WR-2721 could significantly raised the survival rate and suwival time of mice with the combined radiation-burn injury. Prophylactic use of WR-2721 resulted in an increase of LD50 value from 5.19 Gy of the control to 9.53 Gy of the pretreatment. DRF (dose reduction factor) was 1.84. Prophylactic effect of WR-2721 was gained by the immediate use before irradiation with the dose of 200 mg/kg. Pretreatment of WR-2721 in mice with the combined injury brought about a lesser decrease in peripheral WBC count and an earlier recovery compared with the control. Bone marrow myelocyte and monocyte counts were markedly higher than the control at day 1 and 7 postinjury and bone marrow hematopoietic stem cell count was significantly higher than the control at day 7 and 14. Prophylactic effect of WR-2721 on the combined radiation-burn injury was at least partially due to its protective effect on hematopoiesis.
