临床上应用超分割放射治疗疗效不一,为了了解超分割放射的生物学特点,本实验采用200 µm直径HeLa S-3球体为模型,从生长、集落形成率和细胞周期变化(用流式细胞计)等方面比较单次放射,每日一次2 Gy常规放射和两组超分割放射对人体肿瘤细胞的增殖和动力学的影响。,结果:1,在相同总剂量条件下,超分割放射比常规放射和单次放射抑制球体生长作用小。2,球体受放射后由胰酶消化分散成单细胞,其集落形成率以超分割放射组为高。与第一结果相一致,提示在五天疗程中细胞有一定修复和再分布发生。3,细胞周期变化检测表明放射后都有G,2,+M期细胞比例增加,伴以G,1,和S期细胞比例减少。常规放射后G,2, + M期细胞比例较为恒定,而超分割放射后波动大。放射后RNA含量无明显变化。,如采用较大直径内含乏氧细胞的球体为模型,在分割放射期间还可能有再充氧发生,实验结果可能和本实验差异较大。
Controversy exists about the validity of hyperfractionated radiation inverse of conventional fractionated radiation from clinical trials. It is necessary to design a real advantage of hyperfractionated radiation, compared with the conventional fractionation schedule of radiation treatment. To investigate the basic radiobiological response of hyperfractionated radiation, the 200µm HeLa S-3 spheroid were used in this study. The quantitative changes in growth rate, surviving fraction and cell kinetics of spheroids after various dose fraction schedules were compared. The data of growth rate and surviving fraction assays showed that repair and redistribution of cells during the radiation periods are important. Cell kinetics study showed a quite constant pattern of G,2 ,+ M block in the conventional radiaiton group, but a fluctuated pattern was seen in the hyperfractionated radiation groups. For further mimicking the clinical trail, a larger size of spheroid which have certain hyporic cells was necessary to being used in experiment.
