研究骨髓基质细胞对放射损伤复合烧伤小鼠粒单系祖细胞的影响。结果表明:(1)基质细胞培养上清液可支持少量粒单系祖细胞集落(CFU-GM)生长;而骨髓贴壁细胞层对CFU-GM有明显刺激生长作用。(2)加入外源性粒单系集落刺激因子(GM-CSF)时,未见到基质细胞对粒系祖细胞生长的影响。(3)烧伤、放射损伤、放射烧伤复合伤伤后3~5 d,均见到CFU-GM生长受抑制。损伤后仍能见到基质细胞对粒系细胞生长的刺激作用。粒细胞受抑制严重的单放组和复合伤组,基质细胞的刺激作用较单烧伤组明显。以上结果说明,在放烧复合伤造血损伤的修复与重建过程中,骨髓基质细胞起着良好的刺激作用。
The present study was aimed to investigate the effects of bone marrow stromal cell on the growth of granulopoietic progenitors in mice with combined radiation-burn injury. It was found that: (1) The inhibition of CFU-GM growth in the cultured stromal cell was observed, whether extra CSF existed or not, after the 3~5 th day of combined radiation burn injury. Radiation injury and combined injury resulted in marked depression of it, than the burn injury.(2) Stimulation of the cultured stromal cell in the adherent layer on the growth of granulopoietic progenitors could be achieved.This action was obvious when the extra GM-CSF was absent. (3) Stimulation of stromal cells on CFU-GM formation was observed after all 3 kinds of injuryes.The serious suppression of myelopoiesis in radiation and combined injury showed more obvious stimulation of stromal cells. The results indicated that bone marrow stromal cells played a significant role in reconstruction and recovery of hematopoietic disturbance after combined radiationburn injury.
