探讨硫酸镁对大鼠神经干细胞辐射损伤的保护作用。取新生大鼠神经干细胞进行培养,用免疫荧光法进行神经干细胞的鉴定后用含有不同浓度硫酸镁的培养基培养神经干细胞,选择出硫酸镁的合适浓度为0.00125 g/mL。将神经干细胞分为空白对照组、实验对照组(照射)和实验组(照射+硫酸镁)。分别用2和4 Gy 60Co γ射线对实验对照组和实验组进行照射,分别于照射后24、48和72 h用流式细胞仪对各组神经干细胞凋亡率进行检测,同时采用透射电镜对神经干细胞凋亡形态进行观察。与空白对照组比较,实验对照组各时间点神经干细胞的凋亡率明显升高,(t(2Gy, 72h)=30.60,t(4Gy, 72h)=31.85,p<0.05),细胞呈现出明显的凋亡形态,并可见凋亡小体。与实验对照组比较,实验组各时间点神经干细胞的凋亡率明显降低,(t(2Gy, 72h)=9.08,t(4Gy, 72h) p<0.05),且细胞形态损伤明显减轻。硫酸镁能降低电离辐射对神经干细胞的凋亡率,减轻电离辐射所致神经干细胞核损伤的形态。
To explore the neuroprotective effect of magnesium sulfate on the apoptosis of neural stem cells induced by radiation, neural stem cells were obtained from newborn sprague-dawley rats cerebral hemisphere, and the identification of neural stem cell was done by immunofluorescence after culturing for 24 h. Neural stem cells were cultured by different concentrations of magnesium sulfate and the right concentration of magnesium sulfate was chosen to be 0.00125 mg/mL. In the experiment the neural stem cells were randomly divided into 3 groups: blank control group, experimental control group (radiation) and experimental group (radiation and treated with magnesium sulfate). The whole neural stem cells of experimental control group and experimental group were irradiated by a dose of 2 Gy or 4 Gy with γ-rays. The apoptosis were detected at different time after irradiation in the range of 24 h, 48 h, and 72 h, respectively. The CCK-8 and the morphology of neural stem cells were observed by TEM. Compared with blank control group, the apoptosis rate of experimental control group significantly increased (t(2Gy、72h)=30.60, t(4Gy、72h)=31.85, p<0.05). Compared with control group, the apoptosis rate of experimental group significantly decreased(t(2Gy、72h)=9.08,t(4Gy,72h)p<0.05). Accordingly, magnesium sulfate can decrease apoptosis rate and reduce the injury of neural stem cells in the early stage after irradiation.Cited
