采用MTT(Methyl thiazolyl tetrazolium)实验检测姜油树脂对间充质干细胞的细胞毒性;流式细胞术检测细胞中活性氧(ROS)的变化;彗星实验检测细胞DNA受损情况;实时荧光定量PCR(RT-PCR)以及Western Blot检测核因子E2相关因子2 (Nuclear factor-E2-related factor 2, Nrf2)及其下游典型的抗氧化基因(如HO1、NQO1)的表达变化。结果表明,姜油树脂能降低辐射所诱发的活性氧水平,能减少辐射所造成的DNA损伤,并能明显提高发挥辐射防护作用的抗氧化酶、脱毒酶的mRNA及蛋白表达水平。提示姜油树脂可以通过降低辐射诱发的氧化损伤发挥辐射防护作用,其作用机制与Nrf2有关。
The cell toxicity of different concentration of ginger oleoresin on mesenchymal stem cells (MSCs) was judged by MTT(Methyl thiazolyl tetrazolium). Flowing cytometry experiment was taken to observe the change of reactive oxygen species (ROS). Comet assay was used to detect the DNA damage. RT-PCR and Western Blot were used to detect the expression level of nuclear factor-E2-related factor 2 (Nrf2) protein and two typically recognized Nrf2 downstream genes, heme oxygenase-1 (HO-1) and NAD (P) H quinine oxidoreductase 1 (NQO1). The results showed that ginger oleoresin can reduce the level of ROS and the DNA damage of MSCs, induced by irradiation, and increase the expression of HO1 and NQO1 in mRNA and protein levels, which suggests that ginger oleoresin can reduce the oxidative damage of radiation, performing the role of radioprotective.
