为探究JAK-STAT信号通路与炎症因子白细胞介素6、8(interleukn6、8,IL-6、IL-8)是否参与中波紫外线诱导人脐静脉血管内皮细胞(Human umbilical vein endothelial cells, HUVECs)的辐射损伤,使用不同剂量中波紫外线(10、20、40 J/m,2,)照射血管内皮细胞,照射后24 h通过实时荧光定量PCR检测IL-6 mRNA和IL-8 mRNA的表达水平;Western-blot检测JAK1、JAK2、STAT1、STAT3和p-JAK1、p-JAK2、p-STAT1、p-STAT3的表达情况。结果发现,与对照组(0 J/m,2,)相比,细胞经不同剂量紫外线(10、20、40 J/m,2,)照射24 h后,IL-6 mRNA和IL-8 mRNA表达水平均升高,且与受照剂量呈剂量依赖性;JAK1、STAT1、p-JAK1、p-STAT1、p-JAK2、p-STAT3蛋白表达均升高,且上调程度与受照剂量呈剂量依赖性;JAK2和STAT3蛋白表达变化不明显。结果提示,JAK-STAT信号通路及炎症因子IL-6、IL-8参与中波紫外线照射HUVECs细胞损伤的调控。
Human umbilical vein endothelial cells (HUVECs) were irradiated with different doses (10, 20 and 40 J/m,2,) to investigate the role of JAK-STAT pathway and IL-6, IL-8 in UVB irradiation. After 24 h, the expression level of IL-6 mRNA and IL-8 mRNA was detected by real-time fluorescent quantitative PCR, and that of JAK1, JAK2, STAT1, STAT3, p-JAK1, p-JAK2, p-STAT1 and p-STAT3 was measured by Western-blot. Compared with the control group, the expression level of IL-6 mRNA and IL-8 mRNA increased, and that of JAK1, STAT1, p-JAK1, p-STAT1, p-JAK2 and p-STAT3 increased with the dose of UVB, except JAK2 and STAT3. It can be concluded that JAK-STAT pathway and IL-6 together with IL-8 play a role in HUVECs with UVB irradiation.
