To examine the efficiency and fidelity of rejoining of the double strand break(dsb) DNA of the irradiated mouse Waldeyeer s Ring Equivalent (WRE) cells. With lipofectamine or electroporation, the linearized plasmid pPMH16 with two marker genes (neo and gpt)were transferred into the WRE cells from irradiated mice. Through ordinal G418 screen and XHAYM screen, the rejoining efficiency of the broken gpt gene wes evaluated. The rejoining efficiency of gpt gene was 69.75% for control WRE cells, and 36.05% for the WRE cells gotten form the irradiated mice 4h after exposure to 2Gy γ irradiated and 21.50% 4h after exposure to 4Gy γ irradiated. The results showed that the fidelity of the rejoining of dsb of DNA for the irratiated mouse WRE cells was lower than that for control, and as the radiation dose increased,the fidelity decreased. Both the lipofectamine or electroporation could be successfully used in gene transfection for primary culture of mouse WRE cells. And the wrong repair of dsb of DNA in irradiated mouse WRE cells might be caused frequently.
