辐照灭菌可以引起皮肤和骨移植物中胶原和粘多糖的退变,使其生物学稳定性和生物活性降低。选择最适辐照灭菌剂量需要权衡其对组织破坏作用的可容忍水平,与其对微生物杀灭作用的可信赖程度。研究结果说明在尽量减少初始菌量的通常无菌操作条件下,15 kGy是可以同时满足上述两个要求的最适辐照剂量。皮肤和骨移植物的戊二醛预处理,有助于改善辐照引起的真皮和骨基质的生物学不稳定性,有助于减少辐照灭菌皮肤和骨移植物临床应用过程中经常遇到的过度吸收现象。
Radiation sterilization will induce degradation of col lagen and proteoglycans in the skin and bone grafts. Dose choice of radiation steril ization is decided by the balace of the reliable sterilization to the microorganisms the and tolerable destruction to the graft. The present research supports that 15 kGy is optimal dose to fulfill both the above requirements, provided that skin and bone grafts are prepared under sterile working condition with feasible caution to diminish the initial bioburden as far as possible. Glutaraldehyde pretreatment of skin and bone grafts was proposed for overcoming radiation induced degradation of dermal collagen and bone matrix and it is hopeful for restraining the exaggerated resorption of radiation sterilized AAA bones.
