观察了γ射线照射大鼠双侧肾脏对骨代谢的影响。6月龄雄性SD大鼠 20只,随机分为对照组(,n, =10)和双侧肾脏γ射线照射组(,n, =10)。应用,137,Csγ射线照射装置每侧肾脏照射15Gy,剂量率为0.91Gy/min,照射后饲养3个月。收集24h尿,处死动物,收集血、肾脏、腰椎、股骨、胫骨标本。测定血肌酐、尿素氮、钙、磷、25(OH)D,3,、1,25(OH),2,D,3,、PTH和尿蛋白、,β,2微球蛋白、钙、磷、PYD/肌酐等指标。肾脏标本作光镜和透射电镜观察。第1—4腰椎和右侧股骨作骨密度测定,第3腰椎和右侧股骨作成分分析,第4腰椎和左侧胫骨上段作形态计量学分析。结果表明,射线致肾小球、肾小管和肾间质病理性损伤。与假手术组比较,射线照射组血尿素氮、肌酐和尿,β,2微球蛋白分别升高26%(,p<,0.05)、9%和29.6%(,p<,0.05),血清钙降低4%,血清磷升高2%,尿钙增加24%,尿磷减少35%(,p<,0.05),血清碱性磷酸酶活性提高17%(,p<,0.05),血清25(OH) D,3,变化不明显,而1,25(OH),2,D,3,降低54.9%(,p<,0.05),血清PTH升高27.3%(,p<,0.05),尿PYD/肌酐值增加31.7%(,p<,0.05), 腰椎骨密度降低13.0% (,p,<,0.05),第4腰椎骨矿盐含量降低13.1%(,p,<,0.05),右侧股骨骨密度、干重和灰重分别降低3.5%(,P,<,0.05)、7.6%(,p,<,0.05)和11.6%(,p,<,0.05),骨小梁体积、平均骨小梁厚度和结点末端比分别下降13.2%(,p,<,0.05)、18.2%(p,<,0.05)和36.7%(,p,<,0.05),矿化沉积率提高53.9%(,p,<,0.05)。 15Gyγ射线照射大鼠双侧肾脏可致肾功能障碍,并继发以骨转换加速、骨量减少为特点的代谢性骨病。
The work was to observe the effects of γ-rays radiation of rat kidneys on rat bone metabolism. Ten male SD rats aged 6 months were irradiated at their kidneys with 15Gy ,137,Cs γ-rays( 0.91 Gy/min) and were raised for 3 months after the radiation. On collecting 24h urine of rats they were sacrificed for serum, kidney, spine , femur and tibia exams. Results show that theγ-ray irradiation could induce the pathological injuries of renal glomeruli, tubules and mesenchyme. Comparing to the control group, significant chages were found in the irradiated group in terms of their blood urea, nitrogen creatinine, urinal ,β,-2 microglobulin, serum Ca and P, urine Ca and P, activity of serum alkaline phosphatase, 1,25(OH),2, D,3, serum PTH, urine PYD/creatinine, bone mineral density(BMD) of lumbar vertebras, mineral mass of No.4 lumbar vertebra, BMD, dehydrated weight and ash weight of right femur. Marked changes were also found in bone trabecula volum, average bone trabecula thick and the ratio of nodes/points, and rate of mineralizatal deposition. It was concluded that renal dysfunction and metabolic bone disease might occur with the character of accelerated bone turnover and decreased bone mass.
