The membrane filtration method of Bradley and Kohn was improved and used to detect the double strand breaks of DNA in irradiated Chinese hamster ovary cells and mouse bone marrow cells. The percentage of DNA retained on the membrane was inversely proportional to the radiation dose. The method had good reproducibility and was more simple and more sensitive than the classic neutral sucrose gradient centrifugation method. By this method the rejoining of double strand breaks could also be observed in both types of cells. The results provided an additional evidence that the double strand breaks could be repaired in mammalian cells.
