Expression and localization of Smad4, Smad1 and Smad5 in testis of mice exposed to electromagnetic pulses
Changes in expressions of smad4, smad1 and smad5 in the spermatogenic cells of mice after whole-body exposure to electromagnetic pulse (EMP) were studied to elucidate possible mechanisms of the EMP-induced injuries. Twenty adult Balb/c mice were irradiated with 400kV/m EMP to the whole-body. Specimens were harvested on 1, 7, 14, 21and 28 days after the irradiation. Changes in cellular localizations and expressions of Smad4, Smad1 and Smad5 were examined by immunohistochemical ABC method. Quantitative analysis of the immunostaining was made by an image analysis system. In the seminiferous tubules, the localization of Smad4, Smad1 and Smad5 in the EMP group were significantly different from the control group. The expression densities of smad4, smad1 and smad5 changed significantly on 1, 7, 14, 21and 28 days after irradiation. The study indicates that Smad4, Smad1, Smad5 and BMPs/Smad signal pathway may play an important role in the process of impairment and repair of mouse testis irradiated by EMP.
